The case for and against

Updated Oct 17, 2026

Bull case

AgomAb Therapeutics is attractive because ontunisertib has already de-risked its core thesis in ~5 completed studies, with Ph2a STENOVA showing gut-restricted exposure, clean safety/tolerability, and early antifibrotic efficacy in fibrostenotic Crohn’s disease, a setting with ~620K patients across the US, EU5, and Japan and no approved therapies. The case is strengthened by a clear clinical catalyst path in 2H26, when full STENOVA OLE data and Ph2b NOV-ERA can test whether the 12-week structural gains deepen and whether the program can expand into broader IBD and combination use. The stock also appears undervalued at about a ~$660M market cap versus higher-valued peers, while AGMB-447 adds a second shot on goal in IPF with lung-restricted ALK5 biology, clean early safety, and a potentially differentiated add-on profile.

Bear case

AgomAb Therapeutics is viewed negatively because its lead programs remain exposed to the classic risks of clinical-stage biopharma: uncertain safety, uncertain efficacy, and an unclear regulatory path, with the company itself warning that negative safety or efficacy findings could force downward revisions. Although STENOVA showed some improvements in moderate-to-severe, highly refractory FSCD patients, the signals were mixed, the 100 mg QD dose showed minor increases from baseline on centrally read SES-CD, and the S-PRO assessment appeared to hit an efficacy floor after 4-8 weeks, limiting confidence in durability. Fundamentally, the stock also faces a high-cash-burn profile as R&D is expected to ramp and peak in 2033, while the company is still targeting two highly complex, capital-intensive indications with no approved disease-modifying therapy and no certainty of approval or reimbursement.
