The case for and against

Updated Oct 17, 2026

Bull case

Kura Oncology is supported by a compelling combination of early commercial traction and clinically differentiated data, with KOMZIFTI generating 115 new patient starts and more than 250 total prescriptions in 2Q26, plus roughly 40% physician-initiated combination use and strong payer access. The lead asset ziftomenib is showing unusually durable frontline AML efficacy, including 96% CRc in NPM1-m patients, 94% 12-month OS, and central MRD-negativity in 79% of responders at the <0.1% threshold, which meaningfully strengthens the accelerated-approval and backbone-therapy thesis. Financially, the company also has a sizeable $519MM cash position, while the broader pipeline, including KO-7246 and darolifarnib, adds optionality beyond menin inhibition and supports long-term upside.

Bear case

Kura Oncology is viewed pessimistically because ziftomenib’s bear-case profile implies lower-than-expected efficacy in AML, no meaningful safety edge versus competitors, and only a 10% probability of success, which together undermine the program’s commercial value. The company also faces substantial clinical, regulatory, and competitive risk: its pipeline depends on proving efficacy in trials, securing FDA and foreign approvals, and defending tipifarnib with biomarker-based patents that may be harder to identify and enforce than expected. Although Kura reported a Q2 net loss of $68.3M, or ($0.77)/share, and ended the quarter with $519M in cash/equivalents plus $180M in anticipated collaboration payments, it may still need additional financing to fund development and commercialization.
