The case for and against

Updated Oct 23, 2026

Bull case

Kazia Therapeutics is attractive because paxalisib combines brain penetration, once-daily oral dosing, and dual PI3K/mTOR inhibition with a novel nuclear-PI3K/EZH2 mechanism that may help overcome metastasis and checkpoint resistance. Early metastatic TNBC data are especially encouraging, with 6/6 evaluable patients achieving clinical benefit and 5/6 objective responses, alongside biomarker improvements such as reduced circulating tumor cell clusters and exhausted T cells, while no serious adverse events or grade 3+ hyperglycemia were reported. The opportunity is amplified by large addressable markets in TNBC, HR+/HER2- breast cancer, and colorectal cancer, plus additional optionality from GBM and pediatric CNS programs, all of which support asymmetric upside from a modest current enterprise value.

Bear case

Kazia Therapeutics is a high-risk oncology developer whose lead asset, paxalisib, still has to prove durable efficacy, an acceptable safety profile, and clear differentiation across TNBC, HR+ breast cancer, CRC, and GBM. The fundamental bearish case is reinforced by weak clinical precedent for PI3K/AKT/mTOR inhibition in TNBC and GBM, where prior programs have shown limited benefit or failed pivotal endpoints, while the FDA has already said existing GBM data are insufficient for an NDA. With no approved products and ongoing execution, regulatory, and commercial uncertainty, higher development spending and potential dilutive financing could pressure value creation before any meaningful revenue traction emerges.
