The case for and against

Updated Oct 14, 2026

Bull case

ORIC Pharmaceuticals is attractive because its lead programs address large, validated oncology markets with clear mechanistic differentiation: rinzimetostat appears able to reverse AR resistance in mCRPC while enozertinib shows standout CNS activity in EGFR exon 20 insertion and PACC NSCLC. The company’s clinical data support both efficacy and a cleaner tolerability profile, with rinzimetostat showing 6% grade ≥3 TRAEs, zero dose reductions, and no secondary malignancies to date, while enozertinib posted 100% intracranial ORR across measurable CNS disease. Financially, ORIC ended 2Q26 with about $387.6M in cash and marketable securities, and management expects runway into 2H28, giving it funding to reach multiple catalysts.

Bear case

ORIC Pharmaceuticals is a clinical-stage biotech with no commercial revenue, so its valuation depends almost entirely on pipeline execution, especially MEVPRO-1 and the probability that ORIC-944 and ORIC-114 can clear pivotal development and regulatory hurdles. The bearish case is that a readout miss would undermine the mechanistic thesis for PRC2 inhibition in post-abiraterone mCRPC, while the company still faces meaningful safety, efficacy, pricing, and approval risks across its assets. Even with a focused 2025 pipeline prioritization and a 20% workforce reduction in 4Q25, ORIC may still need additional financing, creating dilution risk before any product can generate value.
