The case for and against

Updated Oct 15, 2026

Bull case

Palisade Bio is attractive because PALI-2108 has shown a coherent early pharmacology story across tissue, biomarker, and endoscopic measures, including a 47.5% reduction in SES-CD, a 58.8% drop in fecal calprotectin, and 2/5 patients achieving endoscopic response and remission after 14 days. The drug also demonstrated about 7-fold higher ileal and colonic tissue concentrations than peak plasma levels, sustained PDE4 inhibition at 30 mg QD, and only mild adverse events with no serious discontinuations, supporting the plausibility of a well-tolerated chronic oral therapy. The clearance of ASCENTRA-CD, a cash balance of $125.2M as of mid-2026 funding operations through both Phase 2 primary readouts, and the upcoming 2H27 UC and 1Q28 Crohn’s catalysts further strengthen the fundamental case.

Bear case

Palisade Bio is viewed negatively because its valuation depends on a highly uncertain clinical-stage pipeline, with PALI-2108 assigned only a 40% probability of launch in UC and 30% in CD, and estimated launches not expected until 2030. The company’s DCF framework uses an 11% discount rate and 1% terminal growth rate to support an enterprise value of about $1.8B, but that figure remains fragile given the risk of insufficient efficacy, variable drug activation, and lingering PDE4 tolerability concerns. Its near-term financial cushion is limited, with end-3Q27 cash of roughly $101M against approximately 260M fully diluted shares, while trial delays, competitive IBD therapies, and possible medium- to long-term equity dilution further weaken the outlook.
