The case for and against

Updated Oct 14, 2026

Bull case

Prime Medicine is viewed positively because it has now cleared major regulatory hurdles for two liver-directed programs, PM577a and PM647, while validating its modular prime-editing platform through repeated use of the same LNP delivery system. The Wilson disease program targets the H1069Q ATP7B variant, which represents 30-50% of cases and has preclinical support for biomarker-driven proof of concept, while PM647 showed approximately 53% and 70% hepatocyte editing and restored M-AAT into or above the healthy human range. Its balance sheet and partnered economics also matter: $109 mm of cash funds two programs into 2027, Bristol Myers Squibb provided a $55 million upfront payment plus a $55 million equity investment, and PM359’s RMAT status strengthens the broader curative thesis.

Bear case

Prime Medicine is viewed negatively because its gene-editing pipeline still faces major clinical and regulatory uncertainty, including patient recruitment constraints, off-target editing risks, and no certainty of approval or reimbursement in the US or elsewhere. Its targeted markets in Wilson disease, Alpha-1 antitrypsin deficiency, and cystic fibrosis may prove smaller or more competitive than expected, while pricing and the need to build commercial infrastructure could pressure adoption. Financially, cash of $109 mm as of 30 June versus $191.4M at end-FY25 suggests a high burn profile and likely future capital raises, creating dilution risk and a runway into 2027 that remains an overhang.
