The case for and against

Updated Oct 14, 2026

Bull case

Septerna is well positioned because its Native Complex Platform is producing differentiated oral small-molecule GPCR assets across endocrinology, immunology, inflammation, and metabolic disease, with multiple shots on goal in SEP-479, SEP-631, TSHR, and obesity partnerships. Lead asset SEP-479 has shown an approximately 3- to 4-day half-life that should support once-daily dosing, with Phase 1 data expected in 1Q27 and a strong preclinical package presented at ENDO 2026, while management expects cash to fund operations into at least 2029. The balance sheet is further supported by Novo Nordisk’s $195 million upfront payment and full R&D funding under their agreement, giving Septerna capital to advance programs and absorb clinical risk as it pursues high-unmet-need indications.

Bear case

Septerna is a clinical-stage biotech with no approved products, so value depends heavily on risky pipeline assets such as SEP-631, TSHR, and GLP-1R/GIPR/GCGR rather than current earnings or commercial cash flow. The DCF scenarios still assign only 15% to 30% approval probabilities and rely on 11% discount rates, signaling substantial clinical, regulatory, and partnership uncertainty, especially around Novo Nordisk and U.S.-only sales assumptions. Even under these relatively conservative inputs, the implied firm value of about $1.8B to $2.8B is fragile because setbacks in trials, delayed development, or long-term dilution could quickly undermine the thesis.
